Kommt drauf an.
Normale Signaturen am besten in JPG, transparente Signaturen (z.B. bei einem PopOut Effekt) als PNG, und animierte Sachen als GIF.
Designs und Wallpaper werden meist auch als JPG gespeichert.
Bilder für Templates würde ich persönlich im .png Format speichern, da die Möglichkeit dort mit Interlace zu arbeiten doch sehr praktisch ist. Wenn es sich allerdings um Templates handelt, bei denen es auf Ressourcen ankommt, sollte man jpg's nehmen und soweit herunterrechnen lassen, dass ein optimales Speicherplatz/Qualität-Verhältnis gewährleistet ist.
Wie Fireball angemerkt hat sind von den verbreiteten Formaten nur .png und .gif in der Lage transparenz zu speichern.

Ich speichere meine Avatare & Signaturen IMMER als .png ab, man sagte mir das man diese nicht umbedingt als .jpeg Abspeichern solle und dann hab ich's mir angewöhnt.
wenn du deine signaturen/avas als png abspeicherst kannst du nicht viel falsch machen
bei jpg gibts mehr einstellungen und somit steigt die chance dass du was falsch machst ^^
qualität kann sich nur im jpg format verschlechtern

zwar mach ichs nich immer, aber öfter.
und vorher nie
aber ich seh da selber eigentlich kein riesen unterschied
.png speichert vollkommen verlustfrei, jpeg hingegen ist verlustbehaftet.
Die Unterschiede die daraus entstehen sind allerdings für Leute die Signaturen machen vollkommen irrelevant.
